Electrochemical characterization of Geobacter lovleyi identifies limitations of microbial fuel cell performance in constructed wetlands.

چکیده
Power generation in microbial fuel cells implemented in constructed wetlands (CW-MFCs) is low despite the enrichment of anode electricigens most closely related to Geobacter lovleyi. Using the model representative G. lovleyi strain SZ, we show that acetate, but not formate or lactate, can be oxidized efficiently but growth is limited by the high sensitivity of the bacterium to oxygen. Acetate and highly reducing conditions also supported the growth of anode biofilms but only at optimal anode potentials (450 mV vs. standard hydrogen electrode). Still, electrode coverage was poor and current densities, low, consistent with the lack of key c-type cytochromes. The results suggest that the low oxygen tolerance of G. lovleyi and inability to efficiently colonize and form electroactive biofilms on the electrodes while oxidizing the range of electron donors available in constructed wetlands limits MFC performance. The implications of these findings for the optimization of CW-MFCs are discussed. [Int Microbiol 20(2):55-64 (2017)].

Characterization of the Electric Current Generation Potential of the Pseudomonas aeruginosa Using Glucose, Fructose, and Sucrose in Double Chamber Microbial Fuel Cell
Background: Different concentrations of the simple carbon substrates i.e. glucose, fructose, and sucrose were tested to enhance the performance of the mediator-less double chamber microbial fuel cell (MFC). Objectives: The power generation potential of the different electron donors was studied using a mesophilic Fe (III) reducer and non-fermentative bacteria Pseudomonas aeruginosa</em...
متن کاملPower output and columbic efficiencies from biofilms of Geobacter sulfurreducens comparable to mixed community microbial fuel cells.
It has been previously noted that mixed communities typically produce more power in microbial fuel cells than pure cultures. If true, this has important implications for the design of microbial fuel cells and for studying the process of electron transfer on anode biofilms. To further evaluate this, Geobacter sulfurreducens was grown with acetate as fuel in a continuous flow 'ministack' system i...
متن کاملGraphite electrode as a sole electron donor for reductive dechlorination of tetrachlorethene by Geobacter lovleyi.
The possibility that graphite electrodes can serve as the direct electron donor for microbially catalyzed reductive dechlorination was investigated with Geobacter lovleyi. In an initial evaluation of whether G. lovleyi could interact electronically with graphite electrodes, cells were provided with acetate as the electron donor and an electrode as the sole electron acceptor. Current was produce...
متن کاملCharacterization of microbial current production as a function of microbe-electrode-interaction.
Microbe-electrode-interactions are keys for microbial fuel cell technology. Nevertheless, standard measurement routines to analyze the interplay of microbial physiology and material characteristics have not been introduced yet. In this study, graphite anodes with varying surface properties were evaluated using pure cultures of Shewanella oneidensis and Geobacter sulfurreducens, as well as defin...
متن کاملAnammox enrichment and constructed wetland inoculation for improvement of wastewater treatment performance
This study contributes to the improvement of low-cost biotechnology for wastewater treatment in constructed wetlands (CWs). Constructed wetlands are energy efficient engineered systems that mimic the treatment processes of natural wetlands, removing polluting organic matter, nutrients, and pathogens from water. The aim of this study was to investigate the advisability of the inoculation of hori...
